While significant progress has been made in understanding the effects of the particle sizes of Au and Ag nanoparticles on surface-enhanced Raman scattering (SERS) enhancement, reports on the size effects of copper nanoparticles and copper-related nanoalloys on SERS enhancement remain scarce. In this work, we employed a dual-protectant approach to synthesize highly metallic copper nanoparticles. This method facilitated the production of size-tunable copper nanoparticles, and we investigated their SERS activity by determining the optimal Cu nanoparticle size, balancing strong electromagnetic field enhancement with minimal scattering. Cu nanoparticles of approximately 58 nm exhibited optimal SERS responsiveness to chemical and electromagnetic field enhancements. To achieve compositional tunability of the Cu nanoparticles, we synthesized Au-Cu alloy hollow particles (in the amount of Cu atoms from 29% to 5%) with large particle sizes of 63–75 nm via galvanic replacement/coreduction reactions. Au-Cu nanoparticles smaller than 10 nm were obtained upon subsequent combination with laser ablation, and SERS was downregulated for EM-SERS. Finally, we demonstrated the potential applications of the AuxCuy nanocomposites to activate the Cu-related interfacial catalysis reactions and cellular uptake/exocytosis pathway by using a facile SERS monitor detection.
